WHEATON 227498: Why BOD Bottle Geometry Matters
The WHEATON® 227498 is a
300 mL USP Type I borosilicate glass bottle
designed for biochemical oxygen demand
testing in water and wastewater laboratories.
Its distinctive shoulder and flared-mouth
geometry are intended to reduce air
entrapment and help maintain a water seal
during incubation when a suitable
glass stopper is installed.
The bottle is supplied without a stopper,
without a manufacturer-assigned number
sequence, and without a barcode.
A permanently screened-on writing patch
is provided for laboratory identification.
Critical technical distinction:
The stopper is essential to the intended
stoppered BOD incubation arrangement,
but it is not included
with catalog 227498.
Order and qualify the appropriate
stopper separately.

Understanding Biochemical Oxygen Demand (BOD)
Biochemical oxygen demand describes
the amount of dissolved oxygen
consumed by microorganisms
as they biologically break down
oxygen-demanding material
in a water sample.
In conventional five-day BOD testing,
an initial dissolved oxygen measurement
is compared with a measurement after
the sample or a prepared dilution
has been incubated under
prescribed conditions.
Five-day BOD testing commonly uses
five days of incubation at
20°C in darkness.
The laboratory must apply its approved
method for dilution water, blanks,
seeding, sample preparation,
dissolved oxygen measurement,
and result calculation.
Method distinction:
The oxygen decrease in an incubation
bottle is not automatically the
reportable BOD result for every
sample. Diluted and seeded procedures
require the applicable corrections
and calculations specified by
the laboratory method.
Why BOD Bottle Design Minimizes Air Entrapment
For BOD testing, unwanted oxygen
entering or remaining in the
sample container can interfere
with the intended measurement.
WHEATON addresses this issue
through the geometry of its
BOD bottle family.
| Design Element |
Function |
| Specialized shoulder radius |
Helps sweep trapped air toward the opening during filling |
| Flared bottle mouth |
Provides space to create a water seal around a fitted stopper |
| Compatible tapered glass stopper |
Designed to displace air as the stopper is inserted; sold separately |
| Stopper joint |
Supports use with appropriate BOD and dissolved oxygen measurement equipment |
Proper filling technique, stopper fit,
and inspection for trapped bubbles
remain necessary even with the
specialized bottle design.
The Flared-Mouth Water Seal
WHEATON BOD bottles feature
a flared mouth designed to hold
a water seal around the glass
stopper during incubation.
The water-seal arrangement helps
prevent ambient air from entering
the bottle and influencing the
dissolved oxygen conditions
during the test.
A glass stopper must be correctly
fitted to the bottle before this
arrangement can function as intended.
The stopper is not part of the
227498 purchasing configuration.
Important:
A stopper-free BOD bottle is
not ready for stoppered incubation
without additional components.
Use the laboratory's approved
procedure to establish, inspect,
and maintain the water seal.
Compatible Glass Stoppers - Sold Separately
DWK identifies replacement
USP Type I borosilicate glass
stoppers that fit the WHEATON
60 mL and 300 mL BOD bottle families.
| Stopper SKU |
Style |
Manufacturer Quantity |
| 227670 |
Glass robotic stopper |
12 per case |
| 227672 |
Glass pennyhead stopper |
12 per case |
Both styles belong to WHEATON's
BOD replacement-stopper range.
The selected style should match
the intended handling and
measurement procedure.
Case-quantity planning:
One case of 227498 includes
24 bottles. At one stopper per
bottle, completing all 24 bottles
requires 24 compatible stoppers.
With manufacturer stopper
case quantities of 12,
that corresponds to
two 12-count stopper cases,
assuming the laboratory needs
new stoppers for every bottle.

These are separately cataloged
WHEATON configurations.
The manufacturer publishes different
heights for the stopper-free and
stopper-included products.
Do not use the dimensions or
included-component descriptions
from another SKU for 227498.
Unnumbered Bottles, Barcoding, and Writing Patches
Catalog 227498 is listed as
unnumbered and
not barcoded.
It does, however, include
the manufacturer's large,
permanently screened-on
writing patch.
The writing patch can support
laboratory-applied sample identifiers,
date markings, or other information
required by the laboratory's
sample-tracking process.
The unnumbered configuration
should not be represented as
carrying a factory-applied
serialized number, barcode,
or unique sample identifier.
Five-Day BOD Testing: Application Workflow Overview
WHEATON recommends its 300 mL
BOD bottle format for the
five-day BOD method referenced
in Standard Methods,
21st Edition, Method 5210 B (2005).
A conventional BOD5 workflow
generally includes the following
operations, as specified by
the applicable testing method:
- Prepare the sample, dilution water, blanks, and any required seed or quality-control materials.
- Fill the appropriate BOD bottles carefully to minimize trapped air.
- Measure initial dissolved oxygen according to the approved procedure.
- Install a compatible stopper and establish the required water seal without entrapping air.
- Incubate under the method's specified conditions, commonly five days at 20°C in darkness for BOD5.
- Measure final dissolved oxygen and evaluate the applicable blanks, dilutions, and quality controls.
- Calculate and report the result according to the approved method.
Method caution:
This overview is educational
and does not replace Standard
Methods 5210 B, an applicable
regulatory method, or
the laboratory's approved SOP.
Follow the current method edition
and reporting requirements
adopted by the laboratory.
Common BOD Bottle Handling Issues
| Condition |
Why It Matters |
| Air bubbles after stoppering |
Can change the oxygen conditions in the sample during incubation |
| Incomplete water seal |
Can permit unwanted air exchange during incubation |
| Dirty or damaged bottle |
May affect sample integrity or compromise glassware safety |
| Incorrect stopper |
May prevent a suitable fit or proper sealing arrangement |
| Incorrect incubation conditions |
May invalidate comparisons under the required BOD method |
| Unverified dissolved oxygen probe fit |
Can interfere with measurement setup or handling |
These are practical handling
and quality-control considerations.
They are not quantified product
defect rates or performance claims.
USP Type I Borosilicate Glass Construction
WHEATON identifies its BOD bottle
family as manufactured from
USP Type I borosilicate glass.
Borosilicate glass is commonly selected
for laboratory applications where
chemical durability and resistance
to aqueous attack are important.
The manufacturer's technical
glass reference describes
borosilicate glass types
and their chemical and
physical properties.
However, general properties
of a glass type do not automatically
establish all performance limits
for a finished BOD bottle.
Important distinction:
USP Type I glass does not mean
this bottle is a certified
Class A volumetric measuring device,
supplied sterile, or suitable
for every temperature or pressure
condition. Those attributes
require separate evidence.
Cleaning, Inspection, and Glassware Care
Accurate BOD testing requires
appropriately prepared glassware.
Contamination or residual materials
can contribute to analytical
interference, so cleaning
procedures should be selected
according to the laboratory's
approved BOD method.
DWK's general glassware guidance
recommends careful handling,
suitable non-abrasive cleaning,
thorough rinsing, and inspection
before reuse.
- Inspect before use: Reject bottles with chipped mouths, cracks, or other visible damage.
- Prevent scratching: Avoid abrasive cleaning tools that may weaken the glass surface.
- Remove residues: Follow the BOD method's cleaning requirements and avoid introducing oxygen-demanding contamination.
- Rinse appropriately: Use the rinse-water quality specified by the laboratory procedure.
- Inspect stopper interfaces: Keep the flared mouth and glass-joint surfaces free from damage.
- Maintain traceability: Identify prepared bottles according to laboratory quality procedures.
No catalog-specific validated
autoclaving, depyrogenation,
or sterilization cycle is established
for 227498 in the reviewed
product records.
